Veja a resposta abaixo:

1. Os quantificadores many, much, a lot of, lots of são normalmente empregues da seguinte forma:

afirmativa negativa e interrogativa
substantivos contáveis no plural a lot of (books)
lots of (books)
many (books)
substantivos não-contáveis a lot of (water)
lots of (water)
much (water)

NOTA: lots of é mais informal que a lot of.

Exemplos:

A LOT OF / LOTS OF (afirmativa)
I've got a lot of books. Tenho muitos livros.
He drank a lot of water. Ele bebeu muita água.
I've got lots of magazines. Tenho muitas revistas.
She drank lots of coffee. Ela bebeu muito café.

MANY (negativa e interrogativa)
He didn't buy many books. Ele não comprou muitos livros.
I don't know many people. Não conheço muita gente.
Has he got many books? Ele tem muitos livros?
Did you eat many biscuits? Comeste muitas bolachas?

MUCH (negativa e interrogativa)
I don't drink much water. Não bebo muita água.
I haven't got much time. Não tenho muito tempo.
Did you drink much wine? Bebeste muito vinho?
Have you got much time? Tens muito tempo?

2. Embora normalmente empregues na negativa e interrogativa, many e much podem também ser empregues na afirmativa:

em linguagem formal:
Many children love animals. Muitas crianças adoram animais.
Much has been done to improve the quality of the magazine. Tem-se feito muito para melhorar a qualidade da revista.

com expressões temporais:
He has lived in Portugal for many years. Ele vive em Portugal há muitos anos.

3. Many e much podem igualmente ser empregues a seguir de how, too, so, e as:

HOW
How many cars have you got? Quantos carros tens?
How much money do you have? Quanto dinheiro tens?

TOO
He ate too many biscuits. Ele comeu bolachas a mais.
He drank too much coffee. Ele bebeu café em demasia.

SO
There are so many shops. Há tantas lojas.
There is so much pollution. Há tanta poluição.

AS
You can eat as many biscuits as you like. Podes comer tantas bolachas quanto quiseres.
You can have as much coffee as you like. Podes beber tanto café quanto quiseres.

4. Embora normalmente empregues na afirmativa, a lot of e lots of podem também ocorrer na negativa e interrogativa:

A LOT OF / LOTS OF (negativa e interrogativa)
Were there a lot of / lots of mistakes in your composition? Houve muitos erros na tua composição?
I haven't got a lot of / lots of money. Não tenho muito dinheiro.
