You would say "I am going on vacation" next week ("next week" only as an example here.) or "I took my vacation on the next week."

========
Anyway, if you wish a preposition on the sentence the right one would be "take vacations from my job", yet it could remind your boss of the sentence "take vacations from marriage.", he he.
He/she could think you want a "break" in the "relationship" and maybe it could be suggestive, that is, make him/her guess that you think feel your "relationship" there is satisfactory at all.

Yet, it can be:

I took a vacation from…

EVERYTHING…work, running, dieting, facebook (almost), tumblr, my family, obligations, stress, over-analyzing, insomnia, my routine, boys, training… all of it!

"Alright everybody im taking vacation from my job at the county this week... "

=========
Others ways to say that would be "taking a working vacation" and many others. You can see some of them on the Web, Wordreference etc.

Normalmente,

I usually take (a) vacation from work in the summer.

Mas pode ser, por exemplo,

In my job we take vacations twice a year.
There are no vacations allowed in my job/work.

São sentidos análogos. "From" induz mais a idéia de "sair", ou "se ausentar". "In" serve mais para descrever uma característica do trabalho. Se fosse da escola, por exemplo, faz sentido o uso de "from":

We have vacation from school in december and january.

Semelhantemente,

I took a leave of absence from work/teaching/my team.
I take a short break from studying every 45 minutes.

Pupil, just reminding you again: it´s "vacation" no "s" in it, please.

Just complementing Henry´s very good reply: December and January.

Indeed. But after what they've done to me this past season, Paulo, I really want to decapitate them.
