Downtown is actually a noun, an adverb and an adjective.
Your sentence uses downtown as an adverb, so using in downtown is grammatically incorrect.

AS AN ADVERB:

She lives downtown.
>> Ela mora no centro da cidade.

She lives in the city center (USA)
She lives in the city centre (UK)
>> Ela mora no centro da cidade.
_

AS AN ADJECTIVE:

I work in the downtown area.
>> Eu trabalho na área do centro da cidade / central.

You could say "I work in the downtown area" because in this case downtown is an adjective describing the area.
_

AS A NOUN:

downtown
Definition: the main part of a town or city where there are a lot of stores and offices.

There is a hotel in the heart of downtown.
>> Há um hotel no coração do centro da cidade.
