Gostaria de saber se "up" em "up against" é apenas para enfatizar. Se for, ele pode ser omitido?

Use "up". Não tem que usá-lo nesse contexto, mas soa melhor.

Whom are you playing against? = yes
Whom are you playing up against? = no

What kind of construction problem are you against? = no
What kind of construction problem are you up against? = yes

Note the subtle differences meaning below:
I am against the prohibition of alcohol. (I approve of unrestricted access to alcohol. I want to change the law against alcohol. I do not want alcohol to be restricted.)
I am up against the prohibition of alcohol. (I am competing against the restricted use of alcohol. I cannot achieve a goal I cannot because of the restr4ictions against alcohol.)

Complemento:

"Be up against" é uma expressão idiomática que equivale a "estar enfrentando".
"Be against" equivale a "ser contra, não concordar com: uma idéia, ação, etc." Não implica necessariamente em ação direta como "up against".
